Epigenetic regulators are new popular targets for cancer treatment. One of the promising epigenetic drug targets is Bromodomain-containing Protein 4 (BRD4) that binds to acetylated histone lysine residues and stimulates transcriptional elongation leading to expression of growth-promoting genes.
This protein is evaluated as therapeutic target in myeloid leukemia and several BRD4 inhibitors have shown to be effective against leukemia.
We offer receptor-based BRD4 Inhibitors Targeted Library (377 compounds) based on OTAVAchemicals Drug-like Green Collection. The library has been designed with docking of each compound in binding site of BRD4 X-ray crystal structure (PDB ID: 4J0S). This complex of BRD4 with inhibitor has high resolution 1.84 Å and provides an excellent basis for targeted library preparation.
The compounds have been selected by energy score, presence of hydrogen bond with side chain of Asn140 (this H-bond is present in many complexes BRD4 with inhibitor) and visual inspection for favorable ligand position in binding site. The library comprises drug-like compounds only.
