CREB Binding Protein (CREBBP) is a coactivator of transcription factor CREB that activates genes in cAMP transcriptional pathway. The coactivator binds to transcription factor activation domains positions histone acetyltransferases near specific nucleosomes in target gene promoter regions, recognizing the acetylated lysine residue.
In cancer, the role of CREBBP is not clear and may depend on the physiologic background of the tumor. In prostate cancer, leukemia, melanoma, lung cancer and others CREBBP has oncogenic potential. Several inhibitors of CREBBP have been developed.
We would like to offer you our CREB Binding Protein (CREBBP) Targeted Library (576 compounds in total). To design this receptor-based library we used crystal structure of human CREBBP in complex with dihydroquinoxalinone (resolution 1.10 Å). The bromodomain of CREBBP was used for molecular docking. During the docking, a hydrogen bond constraint between docked ligand and side chain of key residue Asn1168 has been set. To filter molecular complexes obtained with the docking, a scoring function cutoff was used. Finally, the complexes retained after the filtering were subjected to visual analysis in order to discard ligands with improper binding mode.
